Younger Cells, Visibly Younger Skin

The SenoVance RNA product line applies the SenoReverse™ mechanism to skin. It acts on senescent fibroblasts in the dermis and restores their ability to make collagen and maintain the dermal matrix.

The Cells That Keep Skin Firm

Fibroblasts in the dermis make the collagen and matrix that keep skin firm and elastic. As these cells become senescent, that work slows. In the skin of naturally aged mice, treated animals had lower senescence markers than aged controls and longer telomeres, closer to levels seen in young mice.

  1. Senescent Fibroblasts

They lose much of their capacity to make collagen and maintain the dermal matrix.

  2. Function Restored

SenoReverse™ restores cell cycle regulation and function in these cells.

  3. Collagen and Matrix

Collagen synthesis and matrix maintenance are reestablished, improving skin structure and appearance.

  • Skin of naturally aged mice, treated versus controls: SA-β-gal, p21 and CCNG2 were lower and Ki67 higher.
  • Skin telomeres were longer in treated mice than in aged controls by both qPCR and TRF/Southern blot, closer to levels seen in young mice.

A Different Approach to Aging Skin

Fillers add volume, energy-based devices and collagen stimulators remodel tissue, and botulinum toxin relaxes muscle. The SenoVance RNA product line works on the cells that make collagen, restoring their function.

  1. Energy-Based Devices

They use controlled heat to promote tissue and matrix remodeling.

  2. SenoVance RNA Product Line

It acts on senescent fibroblasts and restores their function.

  3. Collagen Stimulators

Materials such as PLLA, PCL and CaHA promote collagen remodeling through a local tissue response.

  4. Fillers

They add volume and structural support.

  5. Botulinum Toxin

It relaxes the muscles behind dynamic wrinkles.

Schematic skin cross-section: energy-based devices act in the dermis; the SenoVance RNA product line acts on fibroblasts in the dermis; collagen stimulators act in the deep dermis; fillers add volume beneath it; botulinum toxin acts on muscle.

Schematic: each approach is shown at its typical depth; layer thicknesses and depths are not to scale.

Results at Day 56

Two separate split-face studies enrolled 86 participants aged 40 to 60, one with a salon protocol and one with a clinic protocol. In both, an independent testing laboratory found that treatment firmed skin and reduced wrinkles at every assessment: days 14, 28, 42 and 56.

  • Two separate split-face studies enrolled 86 participants aged 40 to 60. Of these, 79 completed the day-56 assessments (60 women and 19 men; mean age 48.9 years): 38 with the salon protocol and 41 with the clinic protocol.
  • Inclusion required wrinkles of grade 2 or higher on the Skin Aging Atlas (Asian edition) at all five sites, along with skin laxity.
  • The product was applied to each participant’s right side; the left side remained untreated.
  • Salon protocol: nano-infusion every two weeks (days 0, 14, 28 and 42).
  • Clinic protocol: monthly application after roller microneedling (days 0 and 28), with 0.5 mm needles on the face and 0.2 mm on the neck.
  • Both sides were measured at the same visits on days 0, 14, 28, 42 and 56. Before each measurement, participants acclimated for 30 minutes at 21 ± 1 °C and 50 ± 10% relative humidity. The laboratory did not know which product was being tested.
  • Testing followed Chinese industry association standards for cosmetic anti-wrinkle and firming efficacy. Efficacy required significant improvement from baseline and over the untreated side (P<0.05 for each).
  • Each study tracked 21 instrument measures: 4 with a Cutometer®, 15 with Antera 3D® (wrinkle length, depth and width at five sites) and 2 with VISIA® (wrinkle area). At day 56, all 21 had improved from baseline on the treated side; wrinkle width and area are not shown above.
  • The two studies used different product batches, delivery methods and application frequencies. R0 is a firmness-related parameter; a decrease means firmer skin.
  • The studies measured wrinkles, elasticity and firmness directly; they did not measure biological age.

From Baseline to Day 56

Every site on every participant was measured and photographed at each visit, from baseline to day 56. Wrinkles at every site were significantly reduced, both on visual assessment and on Antera 3D® measurement.

The Studies Continue

Through day 56, each of the 14 measures shown above improved at every visit compared with the one before. Both studies are ongoing: participants continue treatment at the original frequency through month 4, then enter three months of treatment-free follow-up, for about seven months in all.

  • The salon protocol continues every two weeks and the clinic protocol monthly.
  • Monthly assessments with Cutometer, Antera 3D, VISIA and DSLR photography.
